萝卜硫素对免疫抑制小鼠的免疫调节作用

Effects of Sulforaphane on Immunoregulation in Immunosuppression Mice

  • 摘要: 目的 探讨萝卜硫素对免疫抑制小鼠免疫功能的保护作用。方法 实验用小鼠随机分为4组:对照组、环磷酰胺(Cy)模型组、萝卜硫素高剂量组(30 mg/kg)和萝卜硫素低剂量组(15 mg/kg),灌胃给药,比较四组小鼠脾脏指数、胸腺指数、腹腔巨噬细胞吞噬功能、血清溶血素含量以及外周血淋巴细胞转化率;运用RT-PCR半定量法观察免疫调节基因TGF-β和PD-1mRNA的表达。结果 与对照组相比,免疫抑制小鼠血清溶血素OD值(0.702±0.08)、外周血淋巴细胞的转化率(42.22%)、胸腺指数(1.25±0.21)和脾脏指数(2.73±0.26)、吞噬百分率(31.86%)和吞噬指数(0.73±0.06)显著下降(P<0.05);但是与环磷酰胺模型组比较,萝卜硫素组小鼠血清溶血素OD值(1.325±0.10)、外周血淋巴细胞的转化率(51.44%)、胸腺指数(1.89±0.31)和脾脏指数(3.80±0.46)、吞噬百分率(43.56%)和吞噬指数(1.15±0.08)明显增加(P<0.05),免疫调节基因TGF-β和PD-1mRNA的表达量下降,并且具有剂量依赖性。结论 萝卜硫素可明显改善环磷酰胺免疫抑制小鼠的免疫功能,同时能有效抑制免疫调节基因TGF-β和PD-1的过度表达。

     

    Abstract: Objective To investigate the immunoregulation effect of Sulforaphane(SF) in immunosuppression mice. Methods Institute of Cancer Research (ICR) mice were randomly divided into four groups: control group, Cyclophosphamide(Cy) model group, SF high-dose(30mg/kg) group and SF low-dose(15 mg/kg) group. The phagocytosis of macrophage, the content of serum hemolysin, lymphocyte transformation rate, spleen index and thymus index were analyzed. The expressions of TGF-β mRNA and PD-1 mRNA were detected by RT-PCR. Results Compared with control group, Cy-treated mice showed significant decrease in phagocytic percentage(31.86%) and index(0.73±0.06) of macrophage, hemolysin OD value(0.702±0.08), lymphocyte transformation rate(42.22%), spleen index(2.73±0.26), thymus index(1.25±0.21) (P<0.05). Compared with Cy model group, Sulforaphane increased the number of phagocytic percentage(43.56%) and index(1.15±0.08) of macrophage, hemolysin OD value(1.325±0.10), lymphocyte transformation rate(51.44%), spleen index(3.80±0.46), thymus index(1.89±0.31) (P<0.05) and down-regulated the expressions of TGF-β mRNA and PD-1 mRNA in a dose-dependent manner. Conclusion Sulforaphane could improve the immune function in immunodepression mice induced by Cy, and inhibit effectively the overexpression of TGF-β mRNA and PD-1 mRNA.
