Inhibitory Activity of T-VISA-BikDD Plasmid on Tumor Cells in Vitro

  • 摘要: 探讨T-VI SA- B i kDD脂质体对肿瘤治疗的作用。方法 用阳离子脂质体LipofectamineTM2000将T-VISA-BikDD和CMV-BikDD质粒转染入多种肿瘤细胞株中,MTT法检测细胞活性计算死亡率。结果 T-VISA-BikDD质粒经转染进入多种肿瘤细胞,对肿瘤细胞均有不同程度的杀伤作用;同一肿瘤系、不同细胞株间存在不同程度的差异,但存在一定的剂量依赖关系(P<0.01)。结论 T-VISA-BikDD质粒在体外对多种肿瘤细胞具有一定的杀伤作用,能有效地抑制肿瘤细胞的增殖,在临床抗肿瘤基因治疗中有重要意义。

     

    Abstract: Objective To investigate the inhibitory effects of T-VISA-BikDD plasmid on tumor cells in vitro. Methods T-VISA-BikDD and CMV-BikDD plasmids were transfected into a wide variety of tumor cell lines by cationic liposome lipofectamineTM2000.Cell activities were observed by MTT method and cell mortality was analyzed. Results T-VISA-BikDD plasmid demonstrated killing effects in several tumor cell lines. Dose-dependent relationship of lethal effect were observed in the tested tumor cell lines (P<0.01). Conclusion T-VISA-BikDD plasmid has certain lethal effect for a wide variety of tumor cells. It could effectively inhibit the proliferation of tumor cells and shows remarkable implication in clinical antitumor gene therapy.
