Effect of Down-regulated I2PP2A Expression on Proliferation and Migration of Gastric Carcinoma Cell Line BGC-823

  • 摘要: 目的 探讨siRNA沉默I2PP2A基因表达对人胃癌BGC-823细胞增殖和迁移的影响及其可能的分子机制。方法 脂质体转染法将靶向I2PP2A的短发夹状RNA(short hairpin RNA, shRNA)转染BGC-823细胞;Real-time RT-PCR和Western blot观察转染后胃癌BGC-823细胞I2PP2A mRNA和蛋白表达的变化;WST-1法和Transwell小室分别检测各组细胞增殖和细胞迁移能力的变化;最后采用蛋白质印迹分析法检测三组胃癌细胞中cyclin D1和MMP-9蛋白的表达。结果 转染I2PP2A siRNA后胃癌BGC-823细胞的I2PP2A mRNA及蛋白表达明显受抑制;I2PP2AmRNA和蛋白表达下调抑制胃癌BGC-823细胞的增殖和迁移,并降低cyclin D1和MMP-9蛋白的表达。结论 I2PP2A表达下调介导的胃癌细胞增殖抑制和迁移降低可能与cyclin D1和MMP-9表达下调密切相关,I2PP2A可能成为胃癌治疗的新靶点之一。

     

    Abstract: Objective To investigate the inhibitory effect of synthetic I2PP2A short interfering RNA (siRNA) on the proliferation and migration of human gastric cancer cell line BGC-823 and to explore the possible molecular mechanisms. Methods The siRNA targeted I2PP2A shRNA was transfected into BGC-823 cells by LipofectamineTM 2000. Expression of I2PP2A mRNA and protein were examined by Real-time RT-PCR and Western blot, respectively. WST-1 and Transwell chamber were used to observe the proliferation and migration of BGC-823 cells, respectively. The expression of cyclin D1 and MMP-9 protein were also detected by Western blot. Results I2PP2A siRNA significantly inhibited the expression of I2PP2A in gastric cancer cell line BGC-823 at both mRNA and protein levels. Downregulated I2PP2A expression greatly inhibited the proliferation and migration of BGC-823 cells and also down-regulated the expression of cyclin D1 and MMP-9 proteins. Conclusion Down-regulated- I2PP2A-expression-mediated inhibition on the proliferation and migration of gastric carcinoma cells may be closely associated with the decreased expression of cyclin D1 and MMP-9 proteins, indicating a new approach of gene therapy for gastric cancer.
