Abstract:
Objective To investigate the correlation between TLR3c.1377, TLR4Asp299Gly gene polymorphism and EBV infection in EBV-associated gastric carcinoma(EBVaGC) oncogenesis. Methods TLR3c.1377 and TLR4Asp299Gly gene polymorphism were assessed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) in 41 EBVaGCs, 62 EBV-negative gastric carcinoma (EBVnGC) and 64 controls. Finally, the date was analyzed by SPSS. Results (1)There were significant differences in genotype frequency of TLR3c.1377 between gastric carcinoma group and control group (
P=0.025). The T allele frequency of gastric carcinoma group were significantly higher than that of controls (45.6%
vs. 29.7%,
P=0.004). The risk of gastric carcinoma in T allele frequency carriers was significantly higher than that in controls (OR=2.435,
P=0.008). (2)TLR3c.1377 genotype and genotypefrequency were not significantly different among three groups (P>0.05). (3) TLR4Asp299Gly genotypes in groups were all Asp/Asp homozygote. There was no significant difference in genotypes, allele frequency or carrier frequency (P>0.05). Conclusion There is an association between TLR3c.1377 gene polymorphism and gastric crcinoma susceptibility. T allele is a risk factor for gastric carcinoma, while C allele is a protective gene; TLR3c.1377 gene polymorphism has no correlation with EBVaGC susceptibility.
