血浆抗凝血酶Ⅲ、绒毛蛋白1和凝集素含量在维吾尔族妇女宫颈病变进程中的动态变化及意义

Correlation of Dynamic Changes of Candidate Plasma Proteins Expression with Development of Cervical Carcinogenesis in Uyghur Women

  • 摘要: :目的 根据前期工作中运用血浆蛋白质组学研究筛选出的一系列血浆候选差异蛋白指标,探索血浆中AT Ⅲ、VIL1和CLU表达含量在维吾尔族妇女宫颈病变进程中动态变化。方法 收集82例新疆地区维吾尔族宫颈病变血浆样本(宫颈炎22例,CINⅡ~Ⅲ20例,宫颈癌早期20例,宫颈癌晚期20例),用酶联免疫法检测血浆中ATⅢ、VIL1和CLU的表达含量,分析其与宫颈病变进程的关系及相关性。结果 ATⅢ的表达含量随着宫颈病变进程逐渐下降,CLU和VIL1的表达含量随着宫颈病变的进程逐渐升高(P<0.05)。以宫颈炎组作为对照组,ATⅢ、VIL1和CLU的表达含量与CINⅡ~Ⅲ组、宫颈癌早期组比较差异均有统计学意义(P<0.05)。ATⅢ、VIL1和CLU表达含量之间的动态变化关联性分析差异无统计学意义。结论 维吾尔族妇女宫颈癌发生发展与ATⅢ、VIL1和CLU的表达含量动态变化密切相关,可以成为宫颈癌早期诊断的血浆候选预警蛋白标志物。

     

    Abstract: Objective To explore the relationships of the dynamic changes of candidate plasma proteins expression with the development of cervical carcinogenesis in Uyghur women based on the previous plasma proteomic research. Methods We collected 82 cases of plasma samples from Uyghur women, 20 cases with cervicitis (CV), 20 cases with cervical intraepithelial neoplasia (CIN) Ⅱ-Ⅲ, 20 cases with early stage (Ⅰ-Ⅱ) and 20 cases with advanced stage (ⅡB-Ⅳ) cervical squamous cell carcinoma, quantitatively determined the content of plasma proteins ATⅢ, VIL1 and CLU by enzyme linked immunosorbent assay (ELISA) and statistical analysis, and evaluated the specifi city and sensitivity of candidate plasma protein markers. Results With the development of cervicitis to CIN Ⅱ-Ⅲ, early stage or advanced stage cervical cancer, the changes of these plasma proteins altered gradually, with remarkably decrease in the content of ATⅢ, and increase of VIL1 and CLU, with statistical differences (P<0.05). Compared with cervicitis group, there were signifi cant differences of the content of plasma proteins ATⅢ, VIL1 and CLU in CINⅡ-Ⅲ, early stage or advanced stage cervical cancer group (P<0.05). Correlation analysis of these three protein indicators showed no correlation (P>0.05). By the detection of ATⅢ, VIL1 and CLU in the cervical lesions, generally the sensitivity, specifi city and accuracy of the ATⅢ protein were the highest (73.0%, 72.0%, 73.0 %), VIL1 were the next (65.0%, 57.0%, 54.0%), and CLU were the lowest (60.0%, 59.0%, 59.0%). Conclusion The content of plasma proteins ATⅢ, VIL1 and CLU is an important symbol of the process of cervical lesions, and has high predictive value. It is expected to be the early diagnosis of cervical cancer.
