二氢青蒿素对卵巢癌SKOV3细胞的抑制作用

  • 摘要: 目的 研究二氢青蒿素(dihydroartemisinin,DHA)诱导卵巢癌SKOV3细胞凋亡。方法 用MTT法检测DHA对SKOV3细胞的增殖抑制作用,流式细胞仪(FCM)测其凋亡率,反转录-聚合酶链反应(RT-PCR)检测Bcl-2、Bax的表达。结果 双氢青蒿素对卵巢癌SKOV3细胞有明显的增殖抑制效应,呈明显的时间-剂量依赖性。DHA处理SKOV3细胞后,24、28、72 h的IC50值分别为117、35.677、23.382 μmol/L。二氢青蒿素可以抑制SKOV3细胞增殖,促进其凋亡,下调Bcl-2基因的表达,增加Bax基因的表达,并有时间、浓度依赖性。结论 二氢青蒿素对人卵巢癌SKOV3细胞有体外抑制作用, 可能通过下调Bcl-2、增加Bax基因的表达来促进细胞的凋亡。

     

    Abstract: Objective To investigate the effects of Dihydroartemisinin (DHA) on inducing the apoptosis of human ovarian cancer cell SKOV3. Methods The inhibition of DHA on SKOV3 proliferation was detected by MTT. Apoptosis rate was evaluated by fl ow cytometry assay. mRNA expression of Bcl-2 and Bax was analyzed by RT-PCR assay. Results DHA signifi cantly inhibited the proliferation and induced the apoptosis of SKOV3 cells in a time and dose dependent manner. After SKOV3 cells treated with DHA, the IC50 value of 24, 28 and 72 h was 117, 35.677 and 23.382 μmol/L respectively. DHA could inhibit the proliferation and induce the apoptosis of ovarian cancer SKOV3 cells, reduce the expression of Bcl-2 and increase the expression of Bax, in a dose- and time-dependent manner. Conclusion Dihydroartemisinin could be a suppressor for ovarian cancer SKOV3 cells in vitro. DHA could induce the apoptosis of SKOV3 cells by upregulating Bax protein expression and downregulating Bcl-2 gene.
