Detection of p53 H一ras Gene Mutations by PCR-SSCP analysis and Human Papillomavirus Infection in Cervical Carcinoma

Abstract
p53、H-ras gene mutations and human papillomavirus infection were detected in 40 speciments of cervical carcinoma.GP-PCR (general primers-PCR) was applied to testing for various HPV infection.The positive rate was 85 0 o (34/40).The incidence of HPV16 E7 transformed gene was 61·800(21/34)·Otherwise,H-ras、p53 gene mutation were detected by PCR-SSCP analysis (Single strand conformation polymorphism analysis of polymerase chain reaction products).Mutation rates were respectively 22.5%(9/40) and 2.5%(1/40).H-ras mutation was 14.7% (5/34) in 34 HPV-positive,biopsy tissues·The results show:HPV16 E7 transformed gene is a critical gene in the develepment of cervical carcinoma. Mutation of H-ras gene is infrequent in cervical carcinoma. But it is possible to play an important roly in latter stages p53 mutation is infrequent in cervial carcinomal with HPV infection.
