Studies of the Reversion of Malignant Phenotypes and Inhibition of Inavasion of a Highly Metastatic Human Lung Cancer Cell Subline Induced by Sodium Butyrate

Abstract
Highly metastatic human lung cancer cell subline (PGCL3) cells were treated with sodium butyrate in vitro. We found that PGCL3 cells were reversed similar to the phenotypes of normal cell in morphology, proliferation rate, mitotic index,colony—forming ability in semisolid agar culture and Con A agglutination reaction. These results showed that sodium butyrate could inhibited proliferation and induced differentiation of PGCL3 cells.We also found that sodium butyrate treatment could result in significant inhibition of adhesion of cells to laminin substrate, migrative ability through the polycarbonate filter of Boyden chamber,and invasive ability through matrigel. Our studies identified that sodium butyrate could induced differentiation of PGCL3 cells,which was accompanied by decline in invasive ability.
