Abstract
Objective To investigate the expression of t ranscription factor GATA-3 gene in the bone marrow st romal cells (BMSCs) f rom patients with leukemia or normal controls. Methods The expression of GATA-3 gene was analyzed by using RT2PCR2EL ISA in the BMSCs from 34 normal cases and 42 cases with leukemia. The semi-quantitative expression levels of GATA-3 gene in the BMSCs from patients with leukemia were compared with normal cont rols. Results The expression of GATA-3 gene could be detected in the BMSCs from both normal cont rols and the cases with leukemia. The expression rates of GATA-3 gene in the BMSCs from acute myelocytic leukemia (AML ) and chronic myelocytic leukemia (CML) were similar to the normal controls, whereas the expression rate in the BMSCs f rom acute lymphocytic leukemia (ALL) was significant lower than the normal cont rols ( P < 0. 05) . The rank of expression level of GATA-3 gene in the BMSCs was“AML > ALL > normal > CML”. Conclusion The expression of GATA-3 gene could be identified in the BMSCs f rom normal cont rols and patient s with leukemia. It may influence the regulation of hematopoiesis in the BMSCs and the change of expression pattern may play a certain role in pathogenesis and development of leukemia.
