Inhibition Effect of Calcium Channel Antagonist Ni cardipine on Human Malignant Glioma in Serum-free Culture

  • Objective To study the inhibition effect of Nicar-dip ine on U251MG,a human malignant glioma cell line. Methods The growth effects of Nicardipine and EGF on U251MG cultured in serum-free media were observed by MTT colorimeritric analysis. Results In serum-free media EGF significantly stimulated the growth of U251MG i n a dose-dependent manner.The near-maximal effect was seen at 10ng/ml,which w as similar to that of 10% NCS (P>0.05).Nicardipine inhibited U251MG cells proliferation and, even at 1ng/ml,totally blocked the growth-stimulated effect of EGF. Conclusion Nicardipine decreased the growth of U251MG and b locked the effect of EGF,Which may provied a new clue for the treatment of human malignant gliomas.
