Prognostic Impact of GSTM1 and GSTP1 Polymorphism on Gastric Cancer Patients Treated with Platinum-based Adjuvant Chemotherapy

Abstract
Objective To investigate the association of GSTM1 and GSTP1 polymorphisms with the clini2 cal outcome of gast ric cancer patient s t reated with platinum2based adjuvant chemotherapy. Methods A total of 84 patient s with gast ric cancer were recruited. All patient s received platinum2based adjuvant chemotherapy. PCR2LDR (ligation detection reaction) method was used to detect genotypes of GSTM12 null and GSTP1 Ile105Val. Results Of the 84 patient s, the prevalence of the GSTM12null genotype was 51. 2 % and the f requencies of GSTP1 Ile/ Ile, Ile/ Val and Val/ Val genotype were 61. 9 %, 28. 6 % and 9. 7 %, respectively. The recurrence2f ree survival rate and overall survival rate in patient s with GSTP1 Ile/ Val and Val/ Val genotype were significantly higher than those in patient s with Ile/ Ile genotype ( P <0. 05) . Multivariable analysis showed that GSTP1 Ile/ Ile genotype was associated with shorter recur2 rence2f ree survival ( P < 0. 05) . Conclusion Polymorphism of GSTP1 Ile105Val may be a potential prog2 nostic factor in gast ric cancer patient s t reated with platinum2based adjuvant chemotherapy.
