Effects of Sodium Butyrate on Radiosensitivity of Glioblastoma Cell

  • Objective To investigate the effects and mechanisms of sodium butyrate (NaB) on radiosensitivity of glioblastoma cell U-251. Methods Cloning formation assay was used to analyze the effects of NaB to enhance radiosensitivity of U-251 cells. The effects of NaB on the expressions of Ku70 mRNA and protein were detected by RT-PCR and Western blot respectively. Results Cell survival rate was significantly lower and sensitivity enhancing ratio (SER) was 1.23 after the combined treatment. Levels of Ku70 mRNA and protein were suppressed by NaB in a dose-dependent manner. Conclusion Enhanced radiosensitivity of glioblastoma cell U-251 is induced by NaB through an inhibition of Ku70 expression.
