Study on Tumor Growth Inhibition of Chinese Medicine Scutellaria Barbata in Colorectal Carcinoma Model in Nude Mice

Abstract
Objective To observe the inhibition of Scutellaria barbata extract on human colorectal carcinoma model in nude mice and to explore its possible mechanism. Methods The 24 human colorectal cancer nude mice xenograft model were made and were randomly divided into 4 groups:model group,5-Fu group(positive control),Scutellaria barbata low-dose group and Scutellaria barbata high dose group.In order to calculate the inhibition rate,the situation of xenografts in nude mice were observed and recorded in each group.The expression of TWIST and MMP-2 proteins of nude mice transplanted tumor was detected by Western blot and immunohistochemical,and the correlation was analyzed. Results The tumor weight and volume of transplanted tumors of Scutellaria barbata high-dose group was markblely reduced compared with the model group,and there no difference was found between the Scutellaria barbata high-dose group and 5-Fu group (P>0.05).Compared with the model group,the expression of TWIST and MMP-2 proteins in the Scutellaria barbata high dose group was significantly reduced(P<0.05),which was similar to that in the 5-Fu group. Conclusion The inhibition on human colorectal cancer xenograft tumor may be related to the suppression of TWIST and MMP-2 expression of the certain dose Scutellaria barbata.
