Inhibitory Effect of Ginsenoside Rg3 on Growth,Metastasis and Expression of TAMs in Mice Lung Adenocarcinoma

Abstract
ObjectiveTo study the effects of suppressing lung adenocarcinoma growth and metastasis of ginsenoside Rg3 and its mechanism. Methods Twenty-four C57BL/6J mice bearing high metastatic Lewis cells were randomized into three groups: control group,DDP group and Rg3 group. Each group was treated with the appropriate intervention. All the mice were sacrificed by tumor inoculation in 24 days.Lung and subcutaneous tumors were peeled off for histological examination.The metastatic tumor foci on lung surface in mice were observed.Lung metastatic occurring rate and metastatic foci inhibitory rate were counted.CD68 and MVD were determined by immunohistochemistry method with image analysis system. ResultsThe growth inhibition rate in the DDP group and Rg3 group was 39.20% and 54.86%,respectively. Compared with the control group,the weight of tumor in the DDP group and Rg3 group showed a significant difference (P<0.01). The metastatic inhibitory rate in the DDP group and Rg3 group was 30.25% and 58.57%,respectively,which showed significan difference with the control group. great synergetic effect in decreaseing metastatic foci on lung surface. with.The expression of TAMs in the control group,cisplatin group and Rg3 group was (47.21±12.06),(45.67 ±11.90) and (16.11±6.32),respectively; and the expression of MVD were (24.57±4.59),(23.52±4.74) and (14.17± 2.43),respectively. Conclusion Rg3 can produce a synergetic inhibitory activity against the growth and lung metastasis of lung adenocarcinoma. The fact that Rg3 inhibited growth and lung metastasis of lung adenocarcinoma may be associated with its effect of suppressing the expression of TAMs and MVD in tumor tissue.
