Abstract
ObjectiveTo observe the inhibitory effect on oral cancer cell lines KB induced by RNA interference (RNAi) of survivin gene. MethodsChemically synthesized small interference RNA(siRNA) targeting survivin was transfected into KB cells by lipofectamine method. The expression of survivin mRNA and protein were detected by RT- PCR and Western blot respectively. The cell cycle and apoptosis were detected by flow cytometry(FCM).The growth of KB cells was detected by MTT. ResultsThe expression of survivin mRNA and protein were obviously reduced. The cell cycle of KB cells was arrested at G2/M phase.The apoptosis rate was increased and the growth of cells was inhibited. Conclusionsurvivin siRNA can induce apoptosis and reduce proliferation of KB cells by silencing survivin gene expression. RNAi targeting survivin has a potential value in gene therapy of oral cancer.
