Abstract:Objective To study the mechanism of inhibitory effect of ginsenoside SPG-Rg3 on angiogenesis. Methods To observe the effect of SPG-Rg3 on the proliferation and migration of human umbilical vein endothelial cells induced by conditioned medium of B16 melanoma cells (BMCM) by MTT assay, PCNA immunofluorescence staining and migration assay.The VEGF and MMP-9 expression of B16 melanoma cells treated with SPG-Rg3 were analyzed by immunocytochemical staining. Results The promoting effect of BMCM on proliferation and migration of vascular endothelial cells was reduced in SPG-Rg3 groups.Moreover, 5μg/ml SPG-Rg3 inhibited the VEGF and MMP-9 expression of B16 melanoma cells. Conclusion SPG-Rg3 can suppress the promoting effect of B16 melanoma cells on proliferation and migration of vascular endothelial cells by decreasing secretion of VEGF and MMP-9, thus indirectly inhibited tumor angiogenesis.
