Abstract
To establish the human cervical cancer model in nude mice and to investigate its biological features. Methods Human cervical cancer HeLa cells suspension with different concentration was injected into the subcutaneous to establish the human cervical cancer model in nude mice.Success rate of tumor transplantation, general characteristic,tumor growth,metastasis and histological features were observed.The cell growth and apoptosis in tumor tissue were estimated by Flow Cytometry (FCM).The express of the proteins such as Survivin,Caspase-3 and STAT3 in the tumor tissue was measured by the immunohistochemical(IHC) method. Results The tumorigenicity rate is 100%. The transplanted tumors grew well by local invasion without metastasis.The histological appearance of the tumor cells before and after transplantation was very similar. FCM results showed most of the tumor cells are in the proliferation and differentiation period.The xpression of the proteins Survivin,Caspase--3 and STAT3 in the tumor tissue were positive measured by IHC. Conclusion sHuman cervical cancer model in nude mice has been successfully established. Pathological examination, FCM and IHC method for Survivin,Caspase3 and STAT3 proved this animal model has simulated the biological features of human cervical cancer.Our work produced an ideal model for the mechanism research of apoptosis-inducing therapy.The differences in cell growth status from the HeLa cells with different cell suspension concentrations was preliminarily observed.
