Abstract
Objective To study inhibitory effect of TGF-β1 shRNA on human salivary gland mucoepidermoid carcinoma in nude mice submandibular gland and the effect of Ki-67 and VEGF expression. Methods Nude mice submandibular gland were transplanted with Ms cells, Ms cells stably transfected with empty vectors, Ms cells stably transfected with TGF-β1 shRNA, respectively.The tumor volume and weight were measured and observed by growth curve.Immunohistochemistry and immunoflurescence were used to detect the level of Ki-67 and VEGF. Results Compared with Ms cells and Ms transfected with empty vectors, Ms cells stably transfected with TGF-β1 shRNA showed slow growth in tumor volume and weight (P< 0.01), as well as down-regulated the expressions of Ki-67 and VEGF. Conclusion TGF-β1 shRNA could inhibit the tumor growth of Ms cells, the mechanism of which might be related with down-regulation of Ki-67 and VEGF.
