Effects of survivin siRNA on Radiosensitivity of Cervical Carcinoma Cells

  • Objective Survivin, a member of inhibitor of apoptosis protein (IAP) gene family, is expressed in various human cancer tissues, and may facilitate tumor cell evasion from apoptosis, and promote aberrant mitotic progression. This study was to investigate radiosensitivity of human HeLa cell line after blockade of survivin by small interfering RNA (siRNA). Methods mRNA and protein expression levels of survivin were detected by using reverse transcription-polymerase chain reaction (RT-PCR) and Western blot,respectively. Proliferation inhibition rate of hela cells was analyzed by plating colony forming assay. Cell apoptosis was assayed by flow cytometry (FCM). siRNA was transfected into HeLa cells to inhibit the expression of survivin. Results Survivin expression existed in HeLa cells. Survivin siRNA transfection increased cell apoptosis rate and inhibited proliferation to enhance radiosensitivity. Conclusion survivin siRNA could enhance radiosensitivity of HeLa cells.
