Function of Tumor Related Apoptosis Inducing-ligand to Human Osteosarcoma Cells

  • To study the relationship between expression of decoy receptors and susceptibility of human osteosarcoma cells to TRAIL-induced apoptosis. MethodsThe expression of TRAIL receptors in human osteosarcoma cell line MG-63 and fresh osteosarcoma specimens was detected by in situ hybridation and Western blot. Soluble TRAIL segment was expressed in E-coli BL21,refolded and purified. Induction of apoptosis in MG-63 and Jurkat cell was assayed by examination of morphological changes under microscope and qualified by FACS analysis. Cells were counted by Trypan blue and growth curves were drawn. The proliferative activity was assessed by cell division index. ResultsAlthough death receptors were highly expressed and decoy receptors were lowly expressed in MG-63, its apoptosis can′t be induced by TRAIL was detected in it. MG-63 cells became hypertrophical and were ranged in a weaving look under microscope.The proliferation of MG-63 was inhibited by TRAIL. ConclusionIt seems no correlation between expression of decoy receptors and susceptibility of human osteosarcoma cells to TRAIL in MG-63. TRAIL inhibited the proliferation of MG-63 and changed its morphology.
