Abstract
Objective To study the inhibitory effect of antisense VEGF RNA on growth of TE-1 cell in vivo. Methods TE-1 cells t ransfected with VEGF antisense RNA、pcDNA3. 1 plasmid alone were transplanted subcutaneously into nude mice and growth of the t ransfected cells in vivo was investigated. The expression level of VEGF mRNA and protein、MVD were examined in antisense group by RT-PCR analysis and immunohistochemist ry respectively. Apoptotic cell and cell cycle were detected in transfected cells by FCM assay and the electron micrographs. Results Latent period of the tumor formation of the antisense group was lengthened, while body weight 、volume of tumors was significantly smaller than that of empty vector group and control group . The decreased expression level of VEGF protein and mRNA、MVD were found in antisene group by immunohistochemistry. Apoptotic cells were found in antisense group cells. G0 / G1 phase cells were increased and S phase cells were decreased in cell cycle. Conclusions VEGF antisense RNA could inhibit the proliferation of TE - 1 cell in the nude mice, which may apply a useful theory basis for gene therapy of esophageal cancer.
