Abstract
Objective To explore the apoptosis of PANC-1 cells induced by antisense RNA of survivin and the increasing sensitivity effect to 5-Fu. Methods We t ransferred the const ruction recombination antisense RNA of survivin of eukaryotic vector into PANC-1 cells by elect roporation. Expression of survivin mRNA was determined by RT-PCR. Cell Counting and MTT assay were performed to evaluate the sensibility of t ransferred cells to 5-Fu. The rate of apoptosis was tested by FCM. Results Transfection of the antisense RNA of survivin can inhibit the expression of survivin mRNA and inhibit the growth of PANC-1 Cells. The sensitivity of PANC-1 cells to 5-Fu was enhanced after t ransfection . The rate of apoptosis increased af ter antisense RNA of survivin was transferred into PANC-1 cells. Conclusion Antisense RNA of survivin could enhance 5-Fu-induced apoptosis in PANC-1. Combination of Antisense RNA of survivin and 5-Fu could improve the treatment effect of pancreatic cancer.
