THE EFFECT OF~(60)Coγ RAY ON BRAIN PERMEABILITY TO METHOTREXATE IN MICE

  • 摘要: 小鼠头部接受不同剂量60Coγ射线照射后24小时,腹腔注射甲氨喋呤(10mg/kg),半小时后处死动物,采用磷光分析法测定血及脑组织中药物浓度。结果表明单次≥20Gy剂量的照射可使药物入脑浓度显著升高;分割照射亦可不同程度的提高药物的入脑浓度;照射前30分钟给予血卟啉衍化物能增强照射对脑药通透性的影响。

     

    Abstract: This study was designed to evaluate the effects of irradiation on the peameability of mice brain to methotrexate (MTX). MTX was administered by intraperitoneal injection (10mg/kg) to unirradiated mice, and to mice receiving varying doses of cranial irradiation. 30 minutes after injection, MTX concentrations in brain and in blood were detected using phospho reseence method. There was a significant increase of MTX level in brain tissue after a single dose irradiatiOn (≥20Gy). Fractionated irradiation also increased MTX level in brain tissue. The effect of irradiation on brain drug permeability could be enhanced by hematoporphyrin derivative (HPD) given 30 minutes before irradiation.
