紫草素增加小鼠肝癌和Lewis肺癌放射治疗效应的初步研究

INITIAL STUDY ABOUT RABIOSENSITIZING EFFECT OF SHIKONIN FOR MICE HEPATOMA 22 AND LEWIS LUNG CARCINOMA (LLC)

  • 摘要: 本研究观察了紫草素(Shikonin)对小鼠肝癌(Hepatoma 22,H2)和Lewis肺癌(Lewis Lung Carcinoma,LLC)的放射增敏作用。结果发现,肝癌和Lewis肺癌的抑瘤率紫草素加放射组比单纯紫草素或单纯放射高(P<0.05)。紫草素加放射组的肿瘤生长延迟也比单纯紫草素或单纯放射长,此组小鼠的存活时间也较其他组长(P<0.05)。另外,光镜观察到Lewis肺癌紫草素加放射组的肿瘤组织中存在较大坏死灶;超微结构显示在紫草素组和紫草素加放射组的瘤细胞之间有一些巨噬细胞,以紫草素加放射组明显,上述结果说明紫草素对肝癌和Lewis肺癌有一定的放射增敏作用。紫草素加放射可能增强Lewis肺癌小鼠巨噬细胞对瘤细胞的直接杀伤力。带瘤小鼠生存期的延长间接增加了放疗效果。

     

    Abstract: Mice Hepatoma 22 (H22 ) and Lewis Lung Carcinoma (LLC) were used for demonstrating the radiosensitizing effect of shikonin in this study. It was found that for Hss and LLC, the tumor inhibiting rate of shikonin plus radiation group was higher than that by eithor single shikonin or single radiation (P<0.05). And, the tumor growth delay and survival time of tumor-bearing mice of shikonin plus rediation group were also longer than by single shikonia or single radiation (P<0.05). As with LLC, the large necosis was observed in tumor tissue of shikonin ply s radiation group under optical microscope. In addition, ultrastructure observation showed that were some acrophages among tumor cells in shikonin group or shixonin plus radiation group, especially in the latter. The above findings suggest that shikonin has radiosensitizing effect for HZZ and LLC, Beside this, radiation along with shikonin may enhance the capability of macrophages to kill directly tumor cells in LLC, and lengthen the survival time of both Hzz-and LLC-bearjng mice, which may promote indirectly the effect of radiotherapy.
