  • 摘要: 大肠癌细胞株HR-8348用含有100ug/ml,50ug/ml和25ug/ml(藻蓝蛋白1640培养液处理后,用波长为630nm的脉冲铜激光照射细胞12J/cm2,继续培养24小时后用MTT法测得细胞存活率为22.1%,37.6%和89.7%,显示出-定的剂量效应,与市售光敏剂蚕砂叶啉对比观察,证明其杀伤肿瘤细胞的效力优于蚕砂叶啉。实验表明藻蓝蛋白有明显的光敏效果。藻蓝蛋白无毒、无副作用,其粗制品已用于强化营养食品,提纯后应用于肿瘤的激光治疗,可望成为-种有发展前途的光敏剂。

     

    Abstract: The photodynamic effect of the phycocianin excited with a pulsed copper vapour laser on the human colorectal cancer cell line HR8348 was evaluated in our laboratory. The HR8348 cells were incubated in RPMI-1640 medium containing a series concentration of phycocianin (100, 50, 25pg/ ml),and subsequently subjected to an irradiation with a pulsed copper vapour laser with the wave-length of 620nm and energy of 12 J/cm2, 24 hours after irradiation, the survival rates of the cells as determined with MTT assay were 22.1% ,37. 6% and 89. 7写,respectively, demonstrating a linear doseresponsive effect. Compared with the commercially available photosensitizer chlorophyll II derivative CPD4, phycocianin appeared to be more effective in killing the HR8348 cells. With its sat-isfactory nontoxic photosensitivity. phycocyanin would be a refined photosensitizer for the laser ther-apy for cancer.
