The changes of cell cycle during the mucoepidermoid carcinoma cell apoptosis by Doxorubicin

  • 摘要: 在证明10ug/ml阿霉素诱导粘液表皮样癌MEC—1细胞凋亡的基础上,研究细胞凋亡过程中细胞周期变化,以利于临床用药。方法:通过显微镜观察细胞的形态变化和DNA电泳来观察图形,并依靠流式细胞术准确了解用药后细胞周期的变化。结果:光镜下见癌细胞凋亡改变,DNA电泳是典型梯状条图;流式细胞仪测得阿霉素作用5h时,细胞阻于G2期,24h时细胞阻于G1期。结论:阿霉素诱导MEC—1细胞凋亡初期,细胞阻于G2期,后期细胞阻于G1期。

     

    Abstract: We identified that 10ug/ml Doxorubicin can induce the MEC—1 cell apoptosis. And then we studied the changes of cell cycle during the MEC—1 cells apoptosis.These would benefit to the clinic chemotherapy for the mucoepidermoid carcinoma. Methods:we observed the morphological changes of the MEC—1 cell by microscopy after the cells exposing to 10ug/ml Doxorubicin and apoptosis was detected by the DNA gel electrophoresis. We used FCM to study the changes of cell cycle in the MEC—1cell line. Results:The morphological changes of cell apoptosis observed and the DNA gel electrophoresis show the typical apoptotic ladder.The MEC—1 cells arrested in G2 phase at 5h after administration of 10ug/ml Doxorubicin and were blocked in G1 Phase at 24h. Conclusions:In the first time of the MEC—lcells exposing th 10ug/ml YDoxorubicin,the cells arrested in G2 Yphase. Later,the cells were blocked in G1 Phase and induced apoptosis.
