  • 摘要: 目的 研究CDKN2 基因的突变、缺失与原发性胃癌发生发展的关系。方法 用PCR和银染SSCP分析了45 例胃癌组织和22 例相应正常胃粘膜中CDKN2 基因的突变和纯合性缺失情况。结果 发现CDKN2 基因的纯合性缺失率为28-9 %, 其中β型转录子的缺失率为15-6 % ;无一例有CDKN2 基因的突变。结论 胃癌的发生发展与CDKN2 基因纯合性缺失密切相关,与CDKN2基因的突变无关,β型转录子参与维持CDKN2 基因的正常功能,其纯合性缺失与胃癌的发生有关。

     

    Abstract: Objective In order to study the alterations of CDKN2 gene in human gastric cancer. Methods The deletion and mutation of CDKN2 gene in 45 gastric carcinoma and 22 normal gastric tissues were examined using PCR SSCP analysis. Results 28 9%(13/45 cases)deletion were founded in gastric cancer,but no somatic mutation was present. Conclusion The findings suggested that CDKN2 gene deletion play a important role in the gastric tumorigenesis,whereas CDKN2 mutation is absent.
