Effect of Interferon Alpha on Endometrial Adenocarcinoma Cell Line

  • 摘要: 目的: 观察人白细胞干扰素(IFN-α)对我们建立的子宫内膜腺癌细胞系(JEC)的细胞生长和细胞周期的影响。方法: 将不同浓度:2000u/ml、500u/ml、100u/ml的IFN-α与JEC共同培养7天, 检测JEC细胞生长曲线、细胞的凝集素受体表达, 并用流式细胞仪分析细胞周期的变化。结果和结论: IFN-α对JEC细胞的增殖有抑制作用, 浓度越大, 抑制作用越明显, 但不影响凝集素受体的表达; IFN-α可使JEC细胞G1/G/期增加、S期增加和G2期阻滞, 前者与IFN-α浓度呈正相关, 而后者则为负相关。

     

    Abstract: Objective: To observe effect of interferon alpha on cell growth and cell cycles of human endometrial adenocarcinoma cell line (JEC) established in our laboratory . Methods Effect of cell growth curve and lectin receptors on the cells were detected after incubation with different concentration at 2000u/ml, 500u/ml, 100u/ml of interferon alpha for 7 days, and alteration of cell cycle were analysed using flow cytometry. Results and Conclusion: Interferon alpha can inhibit the proliferation of JEC and the growth of the cells were dose-dependently suppressed. There were no changes in expression of lectin receptors. The cells showed G1/G2 and Sincrese, but G2/M arrest, it was positive correlation between the former and the concentration of interferon alpha, however, the two latters were negative correlation.
