Effects on Sensitivity to Chemotherapy in the Raji Cell Transfected with Exogenous HCAP1 Gene

  • 摘要: 目的 探讨外源HCAP1基因产物对B淋巴瘤 /白血病细胞系Raji细胞化疗敏感性的作用。 方法 用脂质体介导法, 把HCAP1基因转染Raji细胞, 经G4 18筛选后, 分别与化疗药物DNR(daunoru bicin, 柔红霉素 )、VP16 (etoposide, 足叶乙甙 )和VCR(vincristine,长春新碱 )联合培养 2 4小时, 用苔盼兰拒染试验计数活细胞。结果 转染HCAP1的Raji细胞, 在不同浓度的DNR(0 .0 1~ 10 .0 μg/ml)和VP16 (5~ 80 μg/ml)作用下, 与转染空载体和未转染对照细胞比较, 细胞活力的抑制作用明显增强 (P <0 .0 5 )、IC50 (半数抑制浓度 )值明显降低 ;但与不同浓度的VCR(12 .5~ 10 0 μg/ml)作用, 细胞活力的抑制和IC50 值与对照细胞相似。结论 HCAP1过表达可明显增加Raji细胞对DNR、VP16作用的敏感性, 但不能显著增加对VCR作用的敏感性

     

    Abstract: Objective To investigate the effects of exogenous HCAP1 gene product on the sensitivity to chemotherapy in B lymphocytic lymphoma/leukemia cell line Raji. Methods HCAP1 gene was transfected into Raji cells by medium with liposome, The Raji cells stably expressing exogenous HCAP1 gene, which were screened by G418, exposed to various concentration of DNR, VP16 and VCR for 24 hours, the cell viability was measured by trypan blue dye exclusion test.
