Clinical Significance of FoxP mRNA Expression in CD4 CD25 Regulatory T Cells Isolated from Patients with Gastric Cancer

  • 摘要: 目的 探讨胃癌患者体内CD4 CD25调节性T细胞的含量及FoxP mRNA在Treg中的表达情况及其临床意义。方法 应用流式细胞术(FCM)测定20例胃癌患者外周血中Treg细胞含量,同时采用RT-PCR技术检测其FoxP mRNA的表达情况,并与20例健康志愿者的外周血进行对比研究和统计学分析。结果 胃癌患者外周血中Treg细胞占CD4 T细胞总数的(12.76±0.46)%,显著高于健康对照组的(6.81±0.66)%,(P〈0.01);FoxP mRNA在胃癌患者外周血Treg细胞中呈现高表达(0.84±0.02)%,明显高于其在健康对照组中表达(0.76±0.03)%,(P〈O.05)。结论 胃癌患者外周血中Treg的数量较正常人明显增高,同时伴随有FoxP,mRNA高表达,提示FoxP3 ,基因可能对Treg有重要的调节功能,从而影响胃癌病人的抗肿瘤自身免疫功能。

     

    Abstract: Objective  To investigate the content s of CD4 CD25 T cells and the expression of FoxP mRNA in these Treg cells in peripheral blood of the patients with gast ric cancer and to characterize their clinical significance. Methods  By using flow cytometry, the levels of CD4 CD25 T cells in peripheral blood of 20 preoperative patient s with gastric cancer were determined. RT-PCR techniques were used to detect the expression of FoxP mRNA in these Treg cells, and the results were analyzed statistically in comparison with that of 20 healthy volunteers. Results  The percentage of CD4 CD25 T cells in total CD4 T cells isolated f rom the patient s with gastric cancer was (12. 76 ±0. 46) %, in comparison, it was (6. 81 ±0. 66) % in those f rom healthy volunteers. It was obviously different between Treg cells amount of gastric patients and that of volunteers ( P < 0. 01) . The FoxP mRNA expression of Treg cells in the patients with gastric cancer (0. 84 ±0. 02) %〕was significantly higher than that in healthy volunteers 〔(0. 76 ±0. 03) %, ( P<0. 05) . Conclusion  The levels of CD4 CD25 T cells in peripheral blood and the FoxP mRNA expression of Treg cells in gast ric cancer patient s were increasing significantly. FoxP genes may play a pivotal role in regulating the function of Treg cells in self-tumor immunity of the patient s with gastric cancer.
