恶性肿瘤染色体端粒行为异常与癌变发生的关系研究

Study on the Relationship between the Behavioral Abnormality of Chromosomal Telomeres inMalignant Neoplasm 's and the Carcinogenesis

  • 摘要: 目的 探讨恶性肿瘤染色体端粒行为异常与癌变发生的关系。方法 粒端联合采用微量全血培养法制备淋巴细胞染色体 ;端粒长度用 Southern杂交法 ;端粒酶活性用 TRAP- PCR法。结果 食管癌、鼻咽癌、肺癌染色体端粒联合率分别为 1 4.38%、35.0 7%、2 4 .40 %, 均高于对照组9.83%、2 0 .2 7%、1 5.60 % (P<0 .0 1 ) ;鼻咽癌、肺癌端粒长度变化分别为伸长 1 6.67%、1 6.67%, 缩短 76.67%、69.0 4 %, 长度无变化 6.66%、1 4.2 9% ;端粒酶活性阳性率为鼻咽癌 82 .1 4%, 肺癌 (非小细胞肺癌 NSCLC 78.3%, 小细胞肺癌 SCLC 1 0 0 % )。结论 端粒联合率的增高、端粒长度的缩短及端粒酶活性表达与细胞癌变密切相关。

     

    Abstract: Objective Exploration of the relationship between the behavioral abnormality of chromosomal telomeres in malignant neoplasm's and the carcinogenesis . Methods The telomeric association was used by peripheral blood lymphocyte cultures, the telomere length was used by means of southern hybridization, the telomerase activity was used by means of TRAP ( telomeric Repeat Amplification protocol )on PCR. Results The telomeric association rate of esophageal carcinoma (14.38% ), Nasopharyngeal Carcinoma NPC (35.07% ), Lung Carcinoma (24.40% ) were found to be higher than that of controls (9.83% 、20.27% and 14.29% ) P <0.01, the telomere length of in NPC and lung carcinoma 16.67% and 16.67% have telomere elongation, 76.67% and 69.04% have telomere reduction, 6.67% and 14.29% exhibited equal lengths as compared with the controls . The positive rates of telomerase activity were 82.14% in NPC, 78.3% in NSCLC 、100% in SCLC. Condusion Increase of the telomeric association retes, the reduction of telomere length and the positive rates of telomerase activity were more closely related to cell carcinogenesis .
