Effect of Arsenic Trioxide on the Human Ovarian Cancer Cell Line SKOV3

  • 摘要: 目的 观察As2O3对人卵巢癌细胞株SKOV3体外生长的影响。方法 采用MTT比色法,测定不同浓度As2O3对SKOV3细胞生长的抑制作用,并绘制浓度一抑制率量效曲线;用倒置显微镜及透射电镜观察药物作用后SKOV3细胞形态变化;流式细胞仪检测细胞周期变化。结果 As2O3对SKOV3有明显抑制作用,且具有浓度和时间依赖性。72h时As2O3的IC50为4.67μM。As2O3 1.0~4.0μM作用于SKOV3细胞在24~72h后均出现G2/M期阻滞,以48h最明显。As2O3 2.0μM作用24h、48h后流式细胞仪检测SKOV3细胞的凋亡率分别为3.68%、6.66%。倒置显微镜和透射电镜均观察到用药后凋亡细胞典型的形态学特征。结论 As2O3对卵巢癌SKOV3细胞的生长有明显的抑制作用,并可诱导SKOV3细胞周期阻滞及细胞凋亡。

     

    Abstract: Objective  To examine the effect of arsenict rioxide (As2 O3>>) on cell proliferation and cell cycle arrest in the human ovarian carcinoma cell line SKOV3. Methods  The SKOV3 cells were cultured in RPMI1640 with different concent ration of As2O3, and harvested in different cultured time. Methyl thiazolyl tetrozolium(MTT) was used to establish the dose-effect curves. Inverted microscope and transmission electron microscope were used to examine the morphologic changes of SKOV3 cells, and cell cycles and apoptosis were examined by flow cytometry ( FCM) . Results  As2O3 could significantly inhibit SKOV3 cells proliferation with a time-and dose-dependent fashion. The SKOV3 cells could be arrested significantly at G2 /M phase by As2O3 . There were typical apoptotic changes in each group under inverted microscope and transmission electron microscope. Conclusion  As2O3 could obviously inhibit the proliferation of SKOV3 cells through inducing apoptosis and S 、G2 / M arrest .
