超抗原SEB诱导人CTL细胞杀瘤条件优化

Optimization for Tumor-cytotoxicity Effect of CTL Induced by Super Antigen SEB

  • 摘要: 目的通过正交设计优化CTL细胞最佳杀瘤条件。方法补体裂解法分离CTL细胞,MTT法测定CTL细胞杀瘤活性,正交设计选择CTL细胞最佳杀瘤条件。结果在SEB浓度为10-6g/ml,SEB作用CTL细胞时间为24h,CTL细胞与肿瘤细胞之比为20∶1情况下CTL细胞杀瘤活性最强。结论CTL细胞最佳杀瘤条件组合为SEB10-6g/ml,作用时间24h,效靶比20∶1。

     

    Abstract: Objective Selecting the best combination of tumor-cytotoxicity effect of CTL by orthogonal design. Methods CTLs were separated by complement lyses method, and then MTT was performed to determine the tumor-cytotoxicity effect of CTL. The optimal combination was obtained by orthogonal design. Results Under the following combination did CTLs reach their full scope on tumor-cytotoxicity, SEB concentration is 10~ -6 g/ml, and the stimulating time is 12 hours, about the ratio of CTLs and tumor cells is 20DK∶1. ...
