Effects of Zoledronic Acid on Osteosarcoma LM8 Cell Line Biological Behaviors

  • 摘要: 目的研究唑来膦酸对骨肉瘤LM8细胞株黏附、迁移、侵袭和基质金属蛋白酶(MMP-2、MMP-9)的影响。方法不同浓度的唑来膦酸处理LM8细胞,用CCK-8法检测细胞毒作用和LM8细胞的黏附力;细胞划痕试验检测LM8细胞的迁移力;transwell小室模型测定LM8细胞对细胞外基质(ECM)的侵袭力;采用酶联免疫吸附实验(ELISA)检测培养上清液中基质金属蛋白酶(MMP-2、MMP-9)的含量。结果经过6.25μmol/L、12.5μmol/L、25μmol/L的唑来膦酸处理后,LM8细胞的黏附、迁移和侵袭力均明显下调,培养上清液中的基质金属蛋白酶(MMP-2、MMP-9)的含量也明显下降。结论唑来膦酸可抑制骨肉瘤LM8细胞的体外黏附、迁移和侵袭力,并同时明显降低LM8细胞分泌基质金属蛋白酶。

     

    Abstract: Objective  To study the effect s of zoledronic acid on adhesion, migration, invasion and expres2 sion of mat rix metalloproteinase (MMP22, MMP29) of osteosarcoma LM8 cell line. Methods  Af ter LM8 cells were t reated with zoledronic acid, Cell Counting Kit28 assay was used to assess LM8 cells ad2 hesion and the effect of zoledronic acid on viability of LM8 cells ; their migration and invasion to ECM were measured by wound assay and t ranswell chamber method respectively. The content s of MMP22 and MMP29 in culture supernatant were detected with EL ISA. Results  At the concent ration of 6. 25μmol/ L, 12. 5μmol/ L, 25μmol/ L, zoledronic acid significantly inhibited LM8 cells adhesion, migra2 tion and invasion to the ECM. In addition, the effect of zoledronic acid on viability of LM8 cells was neg2 ligible. Zoledronic acid significantly inhibited the expression of mat rix metalloproteinase (MMP22, MMP2 9) . Conclusion  Zoledronic acid has anti2adhesion, anti2migration and anti2invasion effect s on osteosarco2 ma LM8 cells in vi t ro. In addition, zoledronic acid can result in the down2regulation of expression of ma2 t rix metalloproteinase (MMP22, MMP29) .
