姜黄素抑制人胃腺癌细胞增殖及对细胞周期影响的研究

Study on Effects of Curcumin on Proliferation Inhibiting and Ce11 Cyc1e of Human Gastric Cancer Ce11s

  • 摘要: 目的 研究姜黄素对人胃腺癌细胞BGC 82 3体外生长及细胞周期的影响。方法 应用倒置显微镜、透射电镜观察细胞形态及超微结构, MTT法检测姜黄素对细胞的增殖抑制率, 流式细胞仪 (FCM )进行细胞周期时相分析。结果 姜黄素可使细胞明显变圆, 体积缩小, 脱壁细胞增多, 以及核浓缩、核碎裂等凋亡特征性改变 ;姜黄素明显抑制人胃腺癌BGC 82 3细胞的增殖, 2 4h、4 8h的IC50 分别为 2 6 .2、2 4 .7μmol/L ;流式细胞仪检测显示姜黄素主要使细胞周期阻滞于S、G2 /M期。结论 姜黄素明显抑制人胃腺癌BGC 82 3细胞的增殖并能诱导凋亡, 其机制可能与细胞的S、G2 /M期阻滞有关。

     

    Abstract: Objective To study the effects of Curcumin on the growth of human gastric cancer cell line BGC-823 and the cell cycle change in vitro. Methods By using inverse microscopy and transmission electron microscope,the morphologic and characteristic ultrastructural alterations of BGC-823 cells were observed;inhibitory rate of cells proliferation was detected by MTT assay in vitro;cell cycle phase was analyzed by flow cytometry.
