尼卡地平抑制胶质瘤生长的体外实验

Inhibition Effect of Calcium Channel Antagonist Ni cardipine on Human Malignant Glioma in Serum-free Culture

  • 摘要: 目的 研究钙拮抗剂尼卡地平(Nicardipine,NC)对 恶性胶质 瘤细胞系U25IMG的细胞生长效应。方法 在无血清培养基中,予不同剂量的NC、EGF及NC和EGF(10ng/ml)干预 ,应用MTT比色法观察它们的细胞生长效应。结果 EGF对U251MG细胞表现出剂量依赖性生长刺激作用,10ng/ml时接近 最大效应,与10%小牛血清的促生长效应相仿(P>0.05);NC抑制U251MG细胞生长,并 完全阻断EGF的效应。结论 尼卡地平能抑制胶质瘤细胞生长并完全阻断EGF的生长刺激作用,为 临床上治疗胶质瘤提供一条新的线索。

     

    Abstract: Objective To study the inhibition effect of Nicar-dip ine on U251MG,a human malignant glioma cell line. Methods The growth effects of Nicardipine and EGF on U251MG cultured in serum-free media were observed by MTT colorimeritric analysis. Results In serum-free media EGF significantly stimulated the growth of U251MG i n a dose-dependent manner.The near-maximal effect was seen at 10ng/ml,which w as similar to that of 10% NCS (P>0.05).Nicardipine inhibited U251MG cells proliferation and, even at 1ng/ml,totally blocked the growth-stimulated effect of EGF. Conclusion Nicardipine decreased the growth of U251MG and b locked the effect of EGF,Which may provied a new clue for the treatment of human malignant gliomas.
