Study of Sensitivity of B Lymphocyte Cell Line from Breast Cancer Patients Carrying a BACR1 Mutation Towards γ Irradiation

  • 摘要: 目的 探索BRCA1+/-乳腺癌患者B-淋巴细胞株对辐射诱变的敏感性,以期为建立肿瘤启动与进展机制研究的离体试验系统做一些初步的探索并提供信息。方法 以60Co处理BRCA1+/--乳腺癌患者淋巴细胞株和正常人淋巴细胞株,在含细胞松弛素B的RPMI1640培养液中培养28小时,利用细胞质阻断微核分析(CBMN),评价二个细胞株对γ射线的敏感性。结果 二个细胞株经辐射处理后,主要的基因组不稳定性指标即双核细胞微核率(MNed BNC)和核质桥率(NPB)均显著升高(P〈0.01~0.05);比较二个细胞株的上述指标,没有发现二者间自发与辐射诱发遗传损伤程度的差异。结论 受试BRCA1+/-淋巴细胞株对辐射诱变的敏感性没有显著改变,在本试验系统下,其基因组稳定性与对照细胞株无明显差异。

     

    Abstract: Objective  The experiment investigated the gamma irradiation sensitivity of B-lymphocyte cell line from breast cancer patient carrying a BRCA1 mutation. The aim was to get some information to establish an in vit ro experiment system for studying the mechanisms associated with tumor initiation and progression. Methods  The cells were exposed 60 Co and followed 28hs post-irradiation culture with RPMI1640 with Cytochalasin B. CBMN was employed to analyze irradiation sensitivity of treated cells. Results  The frequencies of MNed BNC and NPB were significantly increased in both cell lines. No significant differences on above biomarkers with respect to spontaneous and radiation induced damage between tested and cont rol cell line were found. Conclusion  BRCA1 mutation is not very important to chromosomal radiosensitivity. There was no significant decrease of genomic stability in cells with BRCA1 mutation in our in vitro system.
