TNF-α基因修饰联合冷冻法制备的疫苗抗肝癌作用的研究

A study on anti-hepatocarcinoma effect Induced by vaccines made up of cells treated by method of TNF-α gene modifying combined with freezing

  • 摘要: 目的 探索用TNF α基因修饰联合冷冻法制备更高效能的抗肝癌疫苗的可能性。方法 用TNF α基因修饰和冷冻处理的H2 2细胞制成疫苗A, 用TNF α基因修饰未经冷冻处理的H2 2细胞制成苗B ;另外, 用未经TNF α基因修饰行冷冻处理和未行冷冻处理的H 2 2细胞, 分别制备疫苗C和D。比较这 4种疫苗的抗小鼠接种性肝癌的作用。结果 用TNF α基因修饰和冷冻处理H2 2细胞制成的疫苗, 其降低接种性肝癌发生率的作用, 提高脾淋巴细胞IL 2和TNF诱生水平及脾脏CTL对H2 2细胞的杀伤活性的作用, 均明显优于其他 3种疫苗。结论 冷冻处理TNF α基因修饰的H2 2细胞, 可提高该细胞的免疫原性, 从而可提高疫苗的抗小鼠接种性肝癌的效能

     

    Abstract: Objective To investigate the possibility of producing vaccine with higher effect of anti-hepatocarcinoma by means of freezing the tumour cells which have been TNF-α gene modified before. Methods The A and B vaccine were H22 cells modified by TNF-α genes and were treated by means of freezing, or not.
