  • 摘要: 目的 探讨转酮酶样基因TKTL1 在人类结肠癌发生发展中的作用。方法 用实时定量PCR 检测结肠癌及癌旁组织中转酮酶样基因TKTL1 mRNA 表达水平,连续监测法检测各组织中总转酮酶活性。结果 TKTL1 mRNA 在结肠癌组织中比癌旁组织中的表达明显增强,且侵袭性结肠癌比非侵袭性结肠癌表达增强。同样,结肠癌组织中总转酮酶活性比癌旁组织中高( P < 0. 01), 侵袭性结肠癌较非侵袭性结肠癌中转酮酶活性增高( P < 0. 01) 。结论 TKTL1 mRNA 表达与人类结肠癌的发生发展密切相关。

     

    Abstract: Objective  To explore the effect of TKTL1 on occurrence and development of colon cancer. Methods  Real2time PCR was used to determine the mRNA expression of TKTL1 gene in human colon cancer and para2cancer tissue. Continuous monitoring assay was used to determine total t ransketolase ac2 tivity in the human colon cancer and para2cancer tissue. Results  The expression of TKTL1 gene was sig2 nificantly higher in the colon cancer tissues than para2cancer tissue. Further more, the expression of TK2 TL1 gene was significantly up2regulated in the invasive colon cancer compare with the noninvasive colon cancer . The total t ransketolase activity significant increase in the colon cancer tissues compare with para2 cancer tissue. The total t ransketolase activity significant increase in the invasive cancer tissue compare with the noninvasive colon cancer. Conclusion  Expression of TKTL1 mRNA is related to the occurrence and development of human colon cancer.
