LncRNA WT1-AS Knockdown Promotes Invasion and Migration of Triple Negative Breast Cancer MDA-MB-231 Cells

    目的 探讨LncRNA WT1-AS对三阴性乳腺癌细胞MDA-MB-231侵袭和迁移的影响。
    方法 qRT-PCR检测WT1-AS在三阴性乳腺癌细胞中的表达及基因沉默的效率;Transwell实验检测WT1-AS对MDA-MB-231细胞侵袭能力的影响;划痕实验检测WT1-AS对MDA-MB-231细胞迁移能力的影响;Western blot检测WT1-AS对MDA-MB-231细胞E-cadherin、N-cadherin、Vimentin和Snail表达的影响。
    结果 与正常乳腺上皮细胞相比,WT1-AS在三阴性乳腺癌细胞MDA-MB-231、MDA-MB-453、MDA-MB-468中的表达量明显上调;沉默WT1-AS后,MDA-MB-231细胞侵袭和迁移能力明显降低,且E-cadherin表达明显上调,N-cadherin、Vimentin和Snail表达量显著下调。
    结论 WT1-AS可以通过调控EMT促进三阴性乳腺癌细胞MDA-MB-231的侵袭与迁移能力。

     

    Abstract:
    Objective To investigate the effect of LncRNA WT1-AS on the invasion and migration of triple-negative breast cancer MDA-MB-231 cells.
    Methods qRT-PCR was used to detect the expression of WT1-AS and the efficiency of gene silencing in MDA-MB-231 cells. Transwell invasion test and scratch test were used to detect the invasion and migration of MDA-MB-231 cells; Western blot was used to detect the expression of E-cadherin, N-cadherin, Vimentin and Snail in MDA-MB-231 cells.
    Results Compared with normal human mammary epithelial cells, the expression of WT1-AS in MDA-MB-231, MDA-MB-453 and MDA-MB-468 cells were significantly up-regulated. Knockdown of WT1-AS significantly reduced the invasion and migration abilities of MDA-MB-231 cells, the expression of E-cadherin was increased but N-cadherin, Vimentin and Snail expression were decreased.
    Conclusion WT1-AS could promote the invasion and migration of triple-negative breast cancer cells MDA-MB-231 via regulating epithelial-mesenchymal transition.
