Epoxymicheliolide Suppresses Invasion and Migration of U251 Cells by Downregulating MMP-2/9 via NF-κB Signaling Pathway

    目的 探讨环氧木香内酯(EMCL)对胶质瘤U251细胞侵袭和迁移的抑制作用及其机制。
    方法 不同浓度EMCL处理胶质瘤U251细胞48 h后,用CCK-8法检测EMCL对胶质瘤细胞增殖抑制作用及半数抑制浓度IC50;采用划痕实验和Transwell侵袭实验检测EMCL对胶质瘤U251细胞侵袭迁移的影响;采用Western blot检测胶质瘤细胞中MMP-2、MMP-9、p-p65和p65蛋白的表达。
    结果 胶质瘤U251细胞经EMCL处理后,细胞数目显著减少,细胞形态变圆、变小,细胞与细胞间的接触、丝状伪足均显著减少;胶质瘤U251细胞迁移和侵袭能力随着EMCL浓度的升高而显著下降;MMP-2和MMP-9蛋白表达均下调;p-p65/p65的表达下调。
    结论 环氧木香内酯能够抑制胶质瘤U251细胞侵袭和迁移能力。

     

    Abstract:
    Objective To investigate the inhibitory effect of epoxymicheliolide (EMCL) on the invasion and metastasis of glioma U251 cells and related mechanism.
    Methods After glioma U251 cells were treated with different concentrations of epoxymicheliolide for 48 hours, CCK-8 was used to detect the proliferation of glioma cells and IC50; scratch test and Transwell invasion test were used to detect the invasion and migration of glioma U251 cells; Western blot was used to detect MMP-2, MMP-9, p-p65 and p65 protein expression.
    Results After EMCL treatment, the number of U251 cells was significantly reduced, the cell morphology became round and smaller, the cell-to-cell contact and filamentous pseudopods were significantly reduced; the migration and invasion abilities of U251 cells were decreased significantly with the increased concentration of EMCL; MMP-2 and MMP-9 protein expression were down-regulated; p-p65/p65 expression was down-regulated.
    Conclusion Epoxymicheliolide could inhibit the invasion and migration of U251 cells.
