小细胞肺癌中LncRNA AC009336.24的表达及其与化疗耐药的关系

LncRNA AC009336.24 Expression in Small Cell Lung Cancer and Its Relationship with Chemotherapy Resistance

    目的 探讨LncRNA AC009336.24在小细胞肺癌患者中的表达及意义。
    方法 采用实时荧光定量PCR法检测120例SCLC组织、35例癌旁组织及40例正常肺组织中LncRNA AC009336.24的表达,分析LncRNA AC009336.24表达与SCLC患者临床病理特征及化疗敏感度的关系。
    结果 LncRNA AC009336.24在SCLC组织和化疗耐药患者中的表达水平明显高于癌旁组织、正常肺组织和化疗敏感者(均P < 0.001);LncRNA AC009336.24表达与SCLC患者的疾病分期、淋巴结转移、远处转移、化疗耐药相关(均P < 0.05)。高表达LncRNA AC009336.24患者的总生存时间及无进展生存时间均短于低表达者(均P < 0.001)。单因素及多因素分析发现LncRNA AC009336.24表达、疾病分期、远处转移是SCLC患者独立的预后因素(均P < 0.05)。
    结论 LncRNA AC009336.24与SCLC的发生和发展相关,可能是SCLC患者潜在的预后评估生物标志物。

     

    Abstract:
    Objective To investigate the expression of long non-coding RNA AC009336.24 (LncRNA AC009336.24) in small cell lung cancer (SCLC) tissues and its clinical significance.
    Methods The expressions of LncRNA AC009336.24 in 120 cases of SCLC tissues. 35 cases of para-cancerous tissues and 40 cases of normal lung tissues were detected by real-time fluorescent quantitative PCR. The relationship between the expression of LncRNA AC009336.24 and clinicopathological characteristics and chemosensitivity in SCLC patients was analyzed.
    Results The expression level of LncRNA AC009336.24 in SCLC tissues and chemoresistance patients were higher than those in para-cancerous tissues, normal lung tissues and chemosensitive patients (all P < 0.001). LncRNA AC009336.24 expression was positively correlated with disease stage, lymph node metastasis, distant metastasis and chemosensitivity (all P < 0.05). The overall survival time and progression-free survival of the patients with high expression level of LncRNA AC009336.24 were shorter than those with low expression level of LncRNA AC009336.24 (all P < 0.001). LncRNA AC009336.24 expression, disease stage and distant metastasis were independent prognostic factors (all P < 0.05).
    Conclusion LncRNA AC009336.24 is involved in the development of SCLC and may be used as a potential prognostic marker for SCLC.
