Expression and Clinical Significance of LncRNA KCNQ1OT1 in Ovarian Cancer Tissues

    目的 探讨长链非编码RNA KCNQ1重叠转录物1(LncRNA KCNQ1OT1)在卵巢癌组织中的表达及与临床病理特征的关系。
    方法 实时荧光定量PCR法检测卵巢癌组织、癌旁组织及细胞中LncRNA KCNQ1OT1表达, 分析其表达与患者临床病理特征和预后的关系。通过转染siRNA下调LncRNA KCNQ1OT1的表达,检测细胞增殖和侵袭能力的变化。
    结果 卵巢癌组织及细胞株中LncRNA KCNQ1OT1的表达水平显著高于正常卵巢上皮细胞及正常卵巢组织。高表达LncRNA KCNQ1OT1的患者的总生存期(P < 0.001)和无进展生存时间(P < 0.001)较低表达者缩短。LncRNA KCNQ1OT1的表达与FIGO分期、肿瘤分级和远处转移相关。下调LncRNA KCNQ1OT1的表达能够明显抑制细胞的增殖和侵袭。
    结论 LncRNA KCNQ1OT1参与了调节卵巢癌的发生、发展,可能作为卵巢癌患者预后的一个潜在生物标志物和治疗靶点。

     

    Abstract:
    Objective To explore the expression of LncRNA KCNQ1 overlapping transcript 1 (KCNQ1OT1) in ovarian cancer tissues and its relationship with clinicopathological features.
    Methods The expression levels of LncRNA KCNQ1OT1 in ovarian cancer tissues, adjacent normal tissues and ovarian cancer cell lines were assessed by quantitative real-time PCR.Then, the associations of LncRNA KCNQ1OT1 expression levels with clinicopathological features and prognosis were evaluated. In addition, LncRNA KCNQ1OT1 functions in tumor progression and invasion were further determined by siRNA-induced LncRNA KCNQ1OT1 silencing in vitro.
    Results The expression levels of LncRNA KCNQ1OT1 in ovarian cancer tissues and cell lines were significantly higher than those in adjacent non-tumor tissues and normal ovarian epithelial cells. Higher LncRNA KCNQ1OT1 expression levels were associated with a shorter overall survival (P=0.006) and disease-free survival (P=0.001) in ovarian cancer patients. In addition, LncRNA KCNQ1OT1 expression was positively correlated with FIGO stage (P=0.002), tumor grade (P=0.006) and distant metastasis (P < 0.001). Moreover, LncRNA KCNQ1OT1 knockdown in ovarian cancer cells markedly suppressed cell proliferation and invasion.
    Conclusion The LncRNA KCNQ1OT1 is a novel factor involved in ovarian cancer progression, and constitutes a potential prognostic biomarker and therapeutic target for the patients with ovarian cancer.
