肾上腺激素水平对小鼠乳腺癌转移的影响

Effect of Hormones from Adrenal Gland on Breast Cancer Metastasis in Mice

    目的 探讨肾上腺功能水平和肾上腺激素对小鼠乳腺癌转移的影响及机制。
    方法 将小鼠随机分为三组分别为:假手术组、肾上腺移植组和肾上腺切除组, 并接种稳定表达萤光素酶基因的4T1细胞, 观察肿瘤的生长情况并利用印度墨汁染色和小动物活体成像观察肿瘤转移情况;利用伤口愈合实验和Transwell小室实验检测肾上腺素(E)和糖皮质激素地塞米松(DEX)对4T1细胞迁移能力的影响。最后通过RT-PCR, 实时荧光定量PCR和Western blot检测上皮间质化相关分子如E-cadherin、vimentin和ZEB1基因和蛋白水平的表达。
    结果 肾上腺移植组肿瘤生长明显增快, 瘤体最大, 肺转移结节明显增多, 且腹部转移灶最大;而肾上腺切除对肿瘤的生长和转移作用正好相反。DEX明显促进4T1细胞的迁移能力而E作用不明显, 且DEX上调vimentin和ZEB1的表达而下调E-cadherin的表达。
    结论 糖皮质激素促进小鼠乳腺癌的生长和转移, 这一作用可能是通过促进上皮间质化介导的。

     

    Abstract:
    Objective To investigate the effect of the adrenal gland’s function and the adrenal hormone on the breast cancer metastasis and the underlying mechanisms.
    Methods The mice was divided into three groups after adrenal gland transplantation, sham operation and adrenalectomy and then were inoculated with 4T1-luc cells. We monitored the growth and metastasis of tumors using India ink staining and luciferase assay. The effect of epinephrine (E) and dexamethasone (DEX) on 4T1 migration ability was detected by wound healing assay and Transwell assay. And we used RT-PCR, real-time PCR and Western blot to detect the expression of epithelial-mesenchymal transition (EMT) related genes including E-cadherin, vimentin and ZEB1 mRNA and protein.
    Results The tumor in the adrenal gland transplantation group was larger than those in other two groups, and also the pulmonary nodule was increased obviously and the distant metastases in the abdomen were the most significant; while the tumor growth and metastasis showed an opposite effect in the adrenalectomy group. DEX could promote the migration ability of 4T1 cells while epinephrine induced an unobvious effect. Moreover, DEX increased the mRNA and protein expression of vimentin and ZEB1 while decreased E-cadherin expression.
    Conclusion Glucocorticoid promotes the growth and metastasis of the breast tumor in mice, and the effect may be mediated by promoting EMT.
