肺癌患者外周血中MMP-7 mRNA、sMICA、VEGF的表达及其与侵袭转移的关系

Expressions of MMP-7 mRNA, sMICA, VEGF in Peripheral Blood of Lung Cancer Patients and Their Relationships with Invasion and Metastasis

    目的  测定肺癌患者外周血中基质金属蛋白酶-7 mRNA(MMP-7 mRNA)、可溶性主要组织相容性复合体Ⅰ类相关分子A (sMICA)、血管内皮生长因子(VEGF)的表达水平,探讨它们与侵袭转移的关系及临床意义。
    方法  采用荧光定量反转录聚合酶链反应检测110例肺癌患者外周血中MMP-7mRNA表达含量,采用酶联免疫吸附实验检测sMICA、VEGF表达水平,并与40例健康者进行比较。
    结果  肺癌患者外周血中MMP-7 mRNA、sMICA、VEGF表达水平较正常对照组显著升高(P<0.05)。MMP-7 mRNA高表达含量与患者临床分期相关(P<0.05),VEGF高表达水平与病理分型、组织分化程度相关(P<0.05)。肺癌患者外周血中sMICA与MMP-7 mRNA、VEGF的表达水平呈正相关,同时MMP-7 mRNA与VEGF的表达水平呈正相关(P<0.05),治疗有效(包括手术和化疗)的肺癌患者外周血中MMP-7 mRNA、sMICA、VEGF表达水平显著降低(P<0.05),而复发转移患者的表达水平显著升高(P<0.05)。
    结论  MMP-7 mRNA、sMICA和VEGF共同参与了肺癌患者肿瘤转移微环境的构成,与肺癌的发生发展、侵袭转移密切相关。

     

    Abstract:
    Objective  To test the expression levels of matrix metalloproteinases-7 mRNA(MMP-7), solubleMHC class I-related chain A(sMICA), vascular endothelial growth factor(VEGF) in the peripheral blood oflung cancer patients and determine their relationships with the invasion and metastasis and correspondingclinical significance.
    Methods  The expression of MMP-7 mRNA was determined by reverse transcriptionpolymerasechain reaction (RT-PCR) and the levels of sMICA, VEGF were measured with enzymelinked-immunosorbent assay(ELISA). Totally 110 lung cancer patients were regarded as the experimentalgroup and 40 health persons as the control group.
    Results  Compared with the control group, the levelsof MMP-7 mRNA, sMICA and VEGF in the experimental group were significantly increased (P<0.05).The overexpression of MMP-7 mRNA in lung cancer patients were related to clinical stages P<0.05),the high levels of VEGF were related to histopathological types as well as histological grades (P<0.05).The levels of sMICA had a perfect positive correlation with the levels of MMP-7 mRNA and VEGF,meanwhile, the overexpression of MMP-7 mRNA was favorably correlated to the high levels of VEGF in the peripheral blood of lung cancer patients(P<0.05); the levels of MMP-7 mRNA, sMICA and VEGF weresignificantly decreased by effective treatment including surgery and chemotherapy(P<0.05); the increasedexpressions of MMP-7 mRNA, sMICA and VEGF were found in lung cancer patients with recurrence andmetastasis(P<0.05).
    Conclusion  MMP-7 mRNA, sMICA and VEGF which participate in the compositionof the tumor metastasis microenvironment might have an association with occurrence, development, invasionand metastasis of lung cancer.
