Abstract:
Objective To investigate the effects of MNNG on proliferation, cycle and apoptosis of Kazakh normal esophageal epithelial cells. Methods Kazakh normal esophageal epithelial cells were treated with different concentrations of MNNG(0, 0.75, 1.5 and 3 μg/ml).0 μg/ml MNNG was taken as control group. MTT assay was used to measure cell proliferation. Cell cycles and apoptotic rate were detected by flow cytometry and Annexin V-FITC/PI staining. Results After exposed to MNNG(0, 0.75, 1.5 and 3 μg/ml) for 24 h, cell inhibition rates were 0, 12.880%, 28.414%, 44.401% with significant difference between the latter three groups and the control group(P<0.05); Flow cytometry showed that cells ratio in S phase were (26.000± 1.808)%, (29.867±3.769)%, (37.833±3.782)%, (47.100±2.427)% and in G2/M-phase were (10.867±1.747)%, (16.133±1.498)%, 18.533±1.115)%, (14.267±1.848)% with significant difference between the latter three groups and the control group(P<0.05); The rates of apoptotic cells were (0.533±0.252)%, (1.767±0.252)%, (11.033±2.173)%, (26.767±1.955)% with significant difference between the latter three groups and the control group(P<0.05). Conclusion MNNG could significantly inhibit cell proliferation, arrest cell cycles and induce the apoptosis of Kazakh normal esophageal epithelial cells.
