Effect of Exogenous AES on Malignant Behaviors of Colorectal Cancer Cells

  • 摘要: 目的 研究外源性AES(amino-terminal enhancer of split)对结直肠癌(colorectal cancer,CRC)细胞增殖、侵袭及迁移能力的影响。方法 将构建好的pEGFP-aes载体转染入AES表达相对较低的CRC细胞系LoVo(pEGFP-AES)细胞,Cell Counting Kit-8(CCK-8)检测细胞增殖活力;划痕实验检测细胞迁移能力;Transwell检测 AES 对细胞侵袭能力。结果 pEGFP-N1组(转染空载质粒pEGFP-N1的LoVo)AES 可显著抑制LoVo的侵袭和迁移能力(P<0.05),但不影响LoVo的生长及细胞增殖。结论 AES主要通过抑制侵袭迁移能力影响CRC恶性表型。

     

    Abstract: Objective To study the infl uence of amino-terminal enhancer of split(AES) on malignant behaviors of colorectal cancer(CRC). Methods LoVo has a lower expression of AES than other CRC cell lines conserved by our laboratory.The pEGFP-aes construct was transfected into LoVo cells by lipofectamine TM2000 (pEGFPAES). Cell Counting Kit-8 was applied to analyze cell proliferation. The cell invasion and migration abilities were examined by Transwell assay and wound healing assay respectively. Results Contrast to pEGFP-N1(transfect pEGFP-N1 into LoVo cells), AES can significantly repress cancer cells invasion and migration(P<0.05),but had no influence in cell growth and proliferation. Conclusion AES effects CRC's malignancy by repressing its invasion and migration.
