回药爱康方含药血清抑制Lewis肺癌细胞增殖和诱导凋亡的研究

Experimental Research on Effect of Drug-containing Serum of Hui Medicine AiKang Recipe on Proliferation and Apoptosis of Lewis Lung Cancer Cells in vitro

  • 摘要: 目的 观察回药爱康方(HAKF)含药血清对Lewis肺癌细胞(LLC)增殖抑制作用,探讨回药爱康方抗Lewis肺癌的作用机制。方法 采用血清药理学方法,用SD大鼠分为健康对照(NS)组,爱康方低剂量(AKL)组,爱康方中剂量(AKM)组,爱康方高剂量(AKH)组,环磷酰胺(CTX)组,爱康方低剂量联合环磷酰胺(AKL+CTX)组,爱康方中剂量联合环磷酰胺(AKM+CTX)组,爱康方高剂量联合环磷酰胺(AKH+CTX)组,共8组,制备含药血清,用各组含药血清培养LLC细胞,采用MTT法测各组血清对LLC增殖的影响,流式细胞(FCM)Annen V-FITC/PI 标记法检测细胞凋亡率。结果 回药爱康方含药血清能够抑制LLC增殖,且呈剂量和时间依赖性,爱康方高剂量组20%含药血清作用72 h抑制率达26.92%,高剂量联合CTX抑制率最高达54.71%;与对照组比较,各组血清均具诱导LLC细胞凋亡的作用,并与剂量和时间呈正相关,爱康方高剂量联合CTX 作用最强,作用72 h时,细胞凋亡率下降,而坏死率升高。结论 回药爱康方可抑制Lewis肺癌细胞增殖,并诱导其凋亡,爱康方可以增强环磷酰胺对细胞的抑制作用。

     

    Abstract: Objective To study the effect of drug-containing serum of Hui medicine AiKang Recipe on proliferation and apoptosis of Lewis lung cancer cells (LLC) in vitro and its anti-tumor mechanism. Methods A total of 80 male SD rats were divided into 8 groups, low, medium, high dose of medicine AiKang Recipe(AKL\AKM\AKH) groups, cyclophosphamide group(CTX), low, medium, high dose of AiKang Recipe combined with CTX(AKL+CTX\AKM+CTX\AKH+CTX) groups and normal saline(NS) group. The drug-containing serum was prepared by the serum pharmacology method to culture LLC in vitro. The proliferation of LLC was measured by methyl thiazolyl tetrazolium (MTT) method. The cell apoptosis was measured by fl ow cytometry (FCM) and Annexin V-FITC/PI. Results Drug-containing serum of Hui medicine AiKang Recipe had a remarkable growth-inhibition on LLC in vitro in time- and concentrationdependent manners. The inhibition rate(IR) was 26.92% after 20% drug-containing serum in AKH group for 72 h. Inhibition rate was up to 54.71% in high dose of AiKang Recipe combined with CTX. Compared with the control group, each group using drug-containing serum could induce apoptosis of LLC in timeand concentration-dependent manners. AiKang Recipe combined with CTX containing drug serum on LLC for 72h had the strongest effect, ie. apoptosis rate declined while the necrosis increased. Conclusion Hui medicine AiKang Recipe alone or combined with CTX inhibited proliferation and induce apoptosis of LLC.
