E-选择素在食管鳞状细胞癌中的表达及其临床意义

Clinical Signifi cance of E-selectin Expression in Esophageal Squamous Cell Carcinoma

  • 摘要: 目的 探讨食管鳞状细胞癌(简称食管鳞癌)中E-选择素的表达及其临床意义。方法 收集手术切除食管鳞癌及癌旁正常组织标本各58 例,采用实时荧光定量PCR检测E-选择素(E-selectin)mRNA的表达,采用免疫组织化学PV-9000法检测其蛋白的表达,并分析E-选择素表达与临床病理指标的关系。结果 E-选择素mRNA与蛋白在食管鳞癌组织中的表达明显增高,均与食管癌临床分期、淋巴结转移、浸润深度有关(P均<0.05),而与患者性别、年龄、肿瘤部位、病理类型、分化程度、肿瘤大小无显著相关(P均>0.05)。结论 E-选择素mRNA和蛋白在食管鳞癌组织中表达明显增高,与肿瘤的浸润转移密切相关,有望作为判断食管鳞癌疾病进展、预测侵袭转移、评估预后的辅助指标,为食管鳞癌的治疗提供新的生物作用靶点。

     

    Abstract: Objective To explore the clinical signifi cance of E-selectin expression in esophageal squamous cell carcinoma (ESCC). Methods Fifty-eight tissues of ESCC and adjacent normal mucosa each were collected as samples. The expression of E-selectin mRNA was detected by real-time PCR. E-selectin protein levels were determined by immunohistochemical PV-9000. The correlation between E-selectin expression and the clinicalpathological features was analyzed. Results The mRNA and protein expression levels of E-selectin were significantly higher in ESCC than in normal mucosa. There was a positive correlation between the expression of E-selectin and clinical stage, lymph node metastasis and invasion depth (P<0.05). No positive correlation was found between the expression of E-selectin and gender, age, tumor location, pathological type, degree of differentiation or tumor size (P>0.05). Conclusion The expression levels of E-selectin mRNA and protein in ESCC tissues were signifi cantly increased and related with the invasion and lymph node metastasis. The enhanced E-selectin may serve as auxiliary indexes for judging disease progression, invasion, metastasis and prognosis prediction and as a new biological target for the treatment of esophageal squamous cell carcinoma.
