放射性125I 组织间植入对裸鼠人胃癌细胞BGC-823移植瘤侵袭转移的影响

Effects of Radioactive Seeds 125I through Interstitial Implantation on Neoplasm Invasiveness in Xenograft Nude Mice Model of Human Gastric Carcinoma BGC-823

  • 摘要: 目的 观察放射性125I 组织间植入对人胃癌裸鼠移植瘤侵袭的抑制作用,并探讨其可能的作用机制。 方法 将人胃腺癌细胞株 BGC-823接种BALB/C裸鼠右侧腋部皮下建立人胃癌裸鼠移植瘤模型后,随机分为模型对照组(对照组)以及放射治疗实验组(实验组)。对照组不接受125I 组织间植入;实验组分别接受放射性125I 组织间植入30 d、60 d和90 d。通过测定肿瘤体质量、体积及肺转移结节观察其抑瘤作用,采用ELISA测定肿瘤端粒酶含量以及内皮生长因子(vascular endothelial growth factor, VEGF)和碱性成纤维细胞生长因子(basic fibroblast growth factor, bFGF)含量,Western blot法检测磷酸化Erk(p-Erk)1/2蛋白表达。 结果 与对照组比较,放射性125I 组织间植入30 d、60 d和90 d后能够明显减少肿瘤体质量、体积及肺转移结节,降低肿瘤端粒酶以及内皮生长因子和成纤维细胞生长因子含量,同时抑制p- Erk1/2的表达(P<0.05或P<0.01),尤以125I 植入90 d效果最为显著。 结论 对Erk细胞信号转导通路的抑制可能是放射性125I 近距离植入抑制人胃癌裸鼠移植瘤侵袭的机制之一。

     

    Abstract: Objective To study the inhibitory effect of radioactive seeds 125I through interstitial implantation on neoplasm invasiveness, and analyze possible mechanisms. Methods The BGC-823 cells were subcutaneously injected into the right anterior armpit of BALB/C nude mice in order to establish the model of metastatic tumor. The mice of metastatic tumor were randomly divided into control group and experimental group, and mice from the experimental group were treated by interstitial implantation of 125I for 30, 60 and 90 days, respectively. The tumor inhibitory effect was detected by measuring tumor weigh, size and lung metastasis nodules. The levels of telomerase protein in metastatic tumor, VEGF and bFGF in serum were measured by ELISA, and the expression of p-Erk1/2 in tumor was analyzed by Western blot. Results Compared with the control group, tumor weight, size, lung metastasis nodules, and the levels of telomerase protein in metastatic tumor, VEGF and bFGF in serum were significantly decreased due to 125I interstitially implanted. Meanwhile, the expression of p-Erk1/2 in tumor was down-regulated (P<0.05 or P<0.01), especially for 90-day treatment. Conclusion It could be the mechanism that Erk signal pathway in volved in neoplasm invasiveness is inhibited by radioactive seeds 125I through interstitial implantation in xenograft nude mice of human gastric carcinoma.
